General description

β-Estradiol 6-(O-carboxy­methyl)oxime: BSA is an estradiol conjugate and is membrane-impermeable.[1] BSA is linked to the estradiol at position 6 to form the β-estradiol 6-(O-carboxymethyl) oxime: BSA conjugate.[1]

Application

β-Estradiol 6-(O-carboxy­methyl)oxime: BSA has been used:
  • to test its effect on the N-methyl-D-aspartate receptor (NMDAR) currents in female dorsal root ganglion (DRG) neurons[2]
  • to coat microplates for the detection of plasma 17β-Estradiol levels using indirect enzyme-linked immunosorbent assay (ELISA)[3]
  • to test its effect on MutL homolog 1 in colorectal cancer lines[4]

Biochem/physiol Actions

β-Estradiol 6-(O-carboxy­methyl)oxime: BSA mimics estrogen and favors amyloid precursor protein α (sAPPα) secretion in vitro studies with primary rat cortical neurons.[1] It also stimulates mitogen-activated protein kinase (MAPK) activity in human umbilical vein endothelial cells [5]

    1. β-Estradiol 6-((O-carboxy-methyl)oxime: BSA, Product Number E5630, is soluble in PBS to 0.90-1.10 mg/mL, and this will yield a faintly hazy to hazy, colorless to faintly yellow solution. Accordingly, E5630-5MG is soluble in 4.5-5.6 mL of PBS. The actual batch-specific solubility is listed on the corresponding CoA.     1. We have not performed solution stability studies, and have found no information on solution stability in the literature. We would therefore recommend using freshly prepared solutions.

    1. Our method of preparation is considered proprietary information  However, a method of preparation (not necessarily Sigma-Aldrich’s) has been described in J. Biol. Chem., 228, 713-727 (1957).

    1. The Certificate of Analysis has lot specific information for the number of moles of steroid per mole of bovine serum albumin.  From this data and the molecular weights of the components, an approximate molecular weight for the compound can be calculated.  For example, if the moles of beta estradiol to BSA is 32, then the approximate molecular weight for the compound is 76.4 kDa (BSA has a molecular weight of approximately 66.4 kDa and 32 moles of estradiol is approximately 10 kDa more).
