General description

La toxina del cólera es un miembro de la familia de toxinas de subunidad AB5. Consiste en una sola subunidad A responsable de la ADP-ribosilación y cinco subunidades B dispuestas como un anillo pentámero con funciones de unión al receptor de la superficie celular e internalización. La subunidad A cataliza la ADP-ribosilación de la subunidad α de la proteína G estimuladora (Gαs), lo que provoca un aumento de la actividad adenilil ciclasa y los niveles de AMPc. También ADP-ribosila la transducina y la tubulina.

La toxina del cólera y su subunidad A suelen utilizarse para estudiar los mecanismos de transducción de la señal debido a su impacto en la adenilato ciclasa. Además, la toxina del cólera actúa como un adyuvante estimulando las respuestas de los linfocitos B y los linfocitos T auxiliares tipo 2 al inhibir la producción de interleucina-12. La subunidad B no tóxica (CTB) se fija a las células uniéndose al gangliósido GM1, lo que la convierte en una etiqueta útil para las células microgliales en la investigación neurológica. También se ha demostrado que es un excelente marcador para estudios de transporte axónico utilizando métodos inmunohistoquímicos.

Application

La toxina del cólera de Vibrio cholerae se ha utilizado en ensayos de neutralización de toxinas para evaluar la citotoxicidad de las células.
La toxina del cólera procedente de Vibrio cholerae se ha utilizado como control positivo en el análisis del AMPc (AMP cíclico) para enterotoxinas. Se ha añadido como suplemento en medios de cultivo celular de células epiteliales y de tumores primarios.

Biochem/physiol Actions

Toxina que consiste en una subunidad A (27 kDa) rodeada por cinco subunidades B (aproximadamente 12 kDa cada una), que unen la toxina al gangliósido GM1 en la superficie celular.

    1. Dilution of the cholera toxin to 10 ng/mL in water is possible. Storage is recommended at 2-8 °C to avoid freezing, as stability in solution at -20 °C has not been determined.

    1. As listed in the Product Data Sheet, the lyophilized powder must be stored at 2–8°C. The product, as supplied, is stable for 3 years when stored properly.

  5. Hey, Can this product be reconstituted to a solution and administered to mice for systemic upregulation of immune cells?

    1. This product has not been tested for use in animal models, however, the preparation does contain sodium azide as a preservative. Azide is reported to have an LD50 of 19 mg/kg in mouse by I.V. route. This is not a sterile preparation, solutions would require filter sterilization prior to in vitro or in vivo use.     1. Cholera toxin is soluble in water at a concentration of 10 mg/mL. Solutions are reported to be stable for 1 year when stored at 2-8 °C, and will lose biological activity after prolonged exposure to pH below 6 or above 8, according to Mekalanos, J.J., et al., Meth. Enzymology, 165, 169-175 (1988).
