14-3-3 proteins are a multifunctional protein family comprising seven mammalian isoforms that interact with over two hundred different intracellular molecules including kinases, phosphatases, transcription factors, scaffold proteins, and DNA. 14-3-3 proteins are involved in a multitude of biological functions and play a regulatory role in processes such as apoptotic cell death, mitogenic signal transduction, and cell cycle control. 14-3-3 β participates in various processes, among which is the positive regulation of cell cycle progression and tumorigenesis. Over-expression of 14-3-3 β in NIH-3T3 cells confers tumorigenicity in nude mice via activation of the mitogen-activated protein kinase (MAPK) cascade.
