General description

Biotin Conjugated Monoclonal Anti-VEGF Receptor-2 (KDR) (mouse IgG1 isotype) is derived from the KDR-2 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from a BALB/c mouse immunized with a purified recombinant human extracellular VEGF receptor-2 (KDR). Vascular endothelial growth factor (VEGF) is also called vasculotropin (VAS), vascular permeability factor (VPF) and kinase-insert domain containing receptor (KDR8). It is a member of a family of endothelial cell mitogens and angiogenic factors. VEGF is a homodimeric heparin-binding glycoprotein. The KDR gene has been mapped to human chromosome 4q11-q12. The homologous gene for the human KDR gene is mouse Flk1 (fetal liver kinase 1) and rat TKrc.

Specificity

Monoclonal Anti-Vascular Endothelial Growth Factor Receptor-2 (KDR)-Biotin Conjugate antibody is purified from ascites fluid, and conjugated withSulfo-NHS-LC-Biotin. It is specific for human VEGF receptor2. It does not cross react with Flt1, Flt4 or platelet-derived growth factor receptor β (PDGF-R1).
Reacts specifically with human VEGF receptor-2. This clone has also been termed 260.4. It does not recognize Flt1 (VEGFR-1), Flt4 (VEGFR-3), or platelet-derived growth factor receptor (PDGF-R).

Biochem/physiol Actions

Vascular endothelial growth factors receptors (VEGF Rs) are receptor tyrosine kinases that bind to VEGF that plays important role in embryonic vasculogenesis, angiogenesis and homeostasis. VEGF R2 has the ability to form heterodimers with VEGF R1 and VEGF R3. The binding of VEGF to VEGF R2 results in activation of tyrosine amino acids of the cytoplasmic tail of the receptor. This interaction results in the activation of MAPK, PI3K, PKC, FAK and Src kinase pathways. The biological roles of VEGF R2 include in epithelial cell growth, proliferation, survival, migration and vascular permeability and angiogenesis. Due to its role in promoting migration and angiogenesis, VEGF R2 has been implicated in breast and prostate cancer.

Target description

The KDR receptor (VEGFRa), defining marker of hematopoietic stem cells (HSCs), distinguishes pluripotent HSCs from lineage-committed progenitor HSCs.
