Caspase-2 (also known as ICH-1, NEDD-2) is a member of the interleukin-1beta converting enzyme (ICE) family of cystein proteases. Similar as other caspases, caspase-2 also exists in cells as an inactive proenzyme. During apoptosis, procaspase-2 is processed at aspartate residues by self-proteolysis and/or cleavage by upstream caspases. The processed form of caspase-2 consists of large (19 kDa) and small (12 kDa) subunits which associate to form the active enzyme. The active recombinant human caspase-2 was expressed in E. coli. The expressed caspase-2 spontaneously undergoes autoprocessing to yield the subunits characteristic of the native enzyme. The recombinant caspase-2 preferentially cleaves the substrates consisting of consensus sequence VDVAD (e.g., VDVAD-AFC and VDVAD-pNA). The recombinant enzyme can also be used as a positive control in caspase assays or in determining the specificity of caspase substrates.Recommended usage: 0.5-1 unit for fluorometric caspase assays and 2-4 units for colorimetric caspase assays. Optimal working dilutions must be determined by end user.
Unit Definition
Specific Activity: Where one unit of the recombinant caspase-2 is the enzyme activity that cleaves 1 nmol of the caspase substrate VDVAD-pNA (pNA: pnitroanaline) per hour at 37oC in a reaction solution containing 50 mM Hepes, pH 7.2, 50 mM NaCl, 0.1% Chaps, 10 mM EDTA, 5% Glycerol, and 10 mM DTT.
Physical form
Partially purified recombinant caspase 2 with a full length HIS-6-tag. Approximately 10-20% caspase 2 protein. The remainder is E.Coli proteins. Approximately 0.8-2 mg total protein/U of activity; E.Coli proteins have no reactivity. Lyophilized. Reconstitute in 25 μL PBS containing 15% glycerol.
Storage and Stability
Maintain unopened and rehydrated product at -70°C in undiluted, freeze-thaw protected, tightly sealed aliquots. Avoid repeated freeze/thaw cycles. Product stable for 6 months from date of purchased if handled appropriately.
Analysis Note
Partially purified recombinant caspase 3 with a full length HIS-6-tag. Approximately 10-20% caspase 2 protein. The remainder is E.Coli proteins. Approximately 0.8-2 mg total protein/U of activity; E.Coli proteins have no reactivity.
