Specificity

Clone 2L20 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Cyclin T1. It targets an epitope within 23 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the C-terminal half of human Cyclin T1.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Cyclin T1 in HeLa cell lysate.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound with Cyclin T1 peptide at least one hundred thousand-fold (100,000X) higher than with the non-specific control peptide.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected Cyclin T1 in human testis tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Cyclin T1 in HeLa cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Cyclin T1 in human testis tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Cyclin-T1 (UniProt: O60563; also known as CycT1, Cyclin-T) is encoded by the CCNT1 gene (Gene ID: 904) in human. Cyclin-T1 is a member of the cyclin family, which is characterized by the presence of a cyclin box domain. This domain is crucial for binding to cyclin-dependent kinases (CDKs). Cyclin-T1 typically forms a complex with CDK9, which is essential for its function. The structure of Cyclin-T1 includes a Cyclin Box domain that facilitates binding to CDK9, transactivation domain, involved in the regulation of transcription. Cyclin-T1 primarily functions as a regulatory subunit of CDK9, forming the positive transcription elongation factor b (P-TEFb) complex, also plays a role in cell cycle regulation, particularly in the G1/S transition and involved in neuronal differentiation and function. Cyclin-T1 is ubiquitously expressed in various tissues, with higher expression levels in proliferative and differentiating cells and in activated T-cells and macrophages. Cyclin-T1 has been implicated in several diseases due to its role in transcription regulation. During HIV Infection, Cyclin-T1 is hijacked by the HIV-1 Tat protein to enhance viral transcription and replication. Overexpression or dysregulation of Cyclin-T1 can lead to uncontrolled cell proliferation and tumorigenesis. Abnormal Cyclin-T1 activity can contribute to cardiovascular pathologies and neurodegenerative conditions due to its role in gene expression. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
