PVA polymers are functionalized with thiol-reactive groups, which react at a slower rate with thiol groups of crosslinkers (CD/PEG crosslinkers) to form stable hydrogels. The slow gelation kinetics allows enough time to conveniently manipulate the solution before the onset of homogenous gel formation. The thioglycerol is used to maintain hydrogel strength and also as a control to compare cellular behavior in peptide-modified versus unmodified hydrogels.
