About This Item
Application
Human renin is a member of the Peptidase A1 family. It catalyzes the hydrolysis of angiotensinogen to produce angiotensin I. It is used to study human renin-dependent hypertension as well as renin induced obesity .
Biochem/physiol Actions
Renin is a 406 amino acid precursor (45,057 Da) containing a 23 amino acid signal peptide and a 43 amino acid propeptide. Native active renin is a 340 amino acid glycoprotein containing three disulfide bonds. Renin, an aspartyl protease, cleaves angiotensinogen which results in Ang I. Ang I is converted to Ang II by the Ang-converting enzyme (ACE) . Prorenin is hydrolyzed by plasma kallekrein to produce active renin. The human renin transgene is thought to be responsible for increased appetite, obesity, and metabolic changes which are not related to angiotensin II .
Physical properties
Renin is processed as a 406 amino acid precursor (45,057 Da) containing a 23 amino acid signal peptide and a 43 amino acid propeptide. Native active renin is a 340 amino acid glycoprotein containing three disulfide bonds.
Supplied as a solution in 50 mM Tris HCl, pH 8
Unit Definition
One unit will produce one fluorescent unit from the hydrolysis of Renin Substrate I (Arg-Glu(EDANS)-Ile-His-Pro-Phe-His-Leu-Val-Ile-His-Thr-Lys(dabcyl)-Arg) at 37 °C at pH 8.0
Preparation Note
Expressed in mammalian cells as prorenin. The prorenin is activated to renin using trypsin and re-purified.

Renin inhibitors like aliskiren not only block renin but also bind prorenin, thereby inducing a conformational change (like the change induced by acid) allowing its recognition in a renin-specific assay. Consequently, aliskiren can be used to measure prorenin. VTP-27999 is

Some evidence suggests that the direct renin inhibitor aliskiren may increase the risk of severe hyperkalemia, stroke, or acute kidney injury (AKI) when prescribed with angiotensin-converting enzyme inhibitors (ACEi's) or angiotensin-receptor blockers (ARBs). The extent to which concomitant treatment increases

Blood pressure reduction and renin-angiotensin-aldosterone system inhibition are targets for treatment of atherosclerosis. The effect of renin inhibition on coronary disease progression has not been investigated. To determine the effects of renin inhibition with aliskiren on progression of coronary atherosclerosis.

Liquid chromatography tandem mass spectrometry (LC-MS/MS) is increasingly becoming the method of choice for steroid hormone measurements due to small sample volumes, fast analysis times and improved specificity compared to immunoassays. Achievement of demanding analytical targets for steroid analysis is

Hospitalizations for heart failure (HHF) represent a major health burden, with high rates of early postdischarge rehospitalization and mortality. To investigate whether aliskiren, a direct renin inhibitor, when added to standard therapy, would reduce the rate of cardiovascular (CV) death
