General description

Mucin is majorly composed of carbohydrate units, crucial for structural architecture and conformation.[1] The monomer of mucin is 640 kDa, with protein contributing to only 20% of the molecular weight. The carbohydrate component is attached to the threonine and serine side chains.[2] Mucins are classified into secreted and transmembrane mucins.[3]
Mucins or mucus glycoproteins are the main macromolecular components of mammalian mucus.[4] It was also used in a study to evaluate the sputum smears concentrated by cyto-centrifugation for detection of acid-fast bacilli.[5]

Application

Mucin from porcine stomach has been used:
  • in the standard curve generation for quantifying mucin-2 generated during mucosal inflammation in mice[6]
  • as a component of digestion fluid solutions in in vitro human digestion model[7]
  • as a component of simulated saliva fluid[8]

Mucin from porcine stomach was used in studies on the binding site of the galactose-specific agglutinin PA-IL from Pseudomonas aeruginosa.[9]

Biochem/physiol Actions

Mucin has wide pharmacological applications in drug delivery.[1] Mucins play a protective role in inflammation. Deregulation in mucin synthesis contributes to tumorigenesis and chronic inflammation.[3]
Mucus forms the protective cover for all epithelial surfaces. The major structural component of mucus is gel-forming mucins. The mucus layer covering the intestinal epithelium has the MUC2 mucin as its central molecule. MUC2 is produced by goblet cells.[10]

  1. What is the best way to sterilise the product without destroying the structure?

    1. The structural integrity of this product has not been evaluated post-sterilization. A general procedure for sterilizing mucin has been published as follows:
      1. Place dry powdered mucin in a flask.
      2. Cover the mucin with 95% ethanol.
      3. Heat the covered mucin at 70 °C for 24 hours.
      4. Evaporate the alcohol to recover the mucin.

    1. This product has not been tested in comparison to human-source mucin.

  3. I saw that this product is made by peptide degradation. by that way, can it harvest transmembrane mucins? I wonder this product contains transmembrane mucin too, especially MUC1.

    1. This product is a very crude preparation of mucin obtained by digestion of hog stomach with pepsin, followed by precipitation of the mucin with alcohol. The crude mucin is collected by filtration and dried. It is a mixture of glycoproteins. Transmembrane mucins tend to be heavily glycosylated and are more resistant to digestion however, conditions such as pH, temperature, and activity of the enzyme may lead to degradation. Further characterization of the mucins present in the final product is not performed.

    1. No. This product is a crude preparation of mucin; it is a mixture of glycoproteins.  It is obtained by digestion of hog stomach with pepsin, followed by precipitation and other steps.

    1. Product No. M2378 is a crude mucin preparation. Product No. M1778 is a partially purified powder prepared according to the method described in Glenister, D.A. and Salmon. K. Microbial Ecol. in Health & Disease 1, 31, (1988). The amount of bound sialic acids are also different. Please refer to the specification sheets on our website.

  9. How can I solubilize Product M2378, Mucin from porcine stomach?

    1. This crude mucin may not be readily soluble in solution. At 20 mg/mL in 1 M sodium hydroxide, turbid suspensions are observed. To try to increase the solubility, one might try further dilution and sonication, but we have not tried this in our labs.
