General description

17β-hydroxysteroid dehydrogenase type 4 (HSD17B4) gene is mapped to human chromosome 5q2. This gene is of more than 100kbp length. It consists of exons ranging from 21bp to 286bp and it encodes an 80kDa protein.
HSD17B4, also known as D-bifunctional protein (DBP), is highly expressed in liver, heart, prostate and testis. It is a 737 amino acid protein consisting of three functional domains N-terminal 3-hydroxyacyl-CoA dehydrogenase domain, a central 2-enoyl-CoA hydratase domain, and a C-terminal sterol carrier protein 2-like domain.

Application

Biochem/physiol Actions

HSD17B4 (17β-hydroxysteroid dehydrogenase type 4) enzyme plays a major role in β-oxidation of fatty acids and steroid metabolic process. Mutations in HSD17B4 gene lead to Zellweger syndrome and an autosomal recessive disorder called DBP (D-bifunctional protein) deficiency, both of which are generally fatal. Increase in the expression of peroxisomal protein leads to prostate cancer.
