General description

Laminin protein is a non-collagenous glycoprotein isolated from Engelbreth-Holm-Swarm (EHS) mouse sarcoma. A laminin protein molecule is composed of three disulfide-linked polypeptides that assemble to form an asymmetrical cross-shaped structure. Purified laminin mouse purified promotes the adhesion and growth of human carcinoma and sarcoma cells, human retinoblastoma cells, liver cells, mouse neuroblastoma cells, mouse embryonal carcinoma cells, and seems to be involved in the development of embryonal tissue. The optimal laminin concentration may vary with the cell type and has to be determined experimentally.

Application

The purified mouse laminin has been used as a coating substrate for culturing and maintaining mouse embryonic stem cell-derived neuronal cultures, providing a suitable surface for cell attachment and growth during in vitro experiments.

Biochem/physiol Actions

Laminin proteins are essential components of the basement membrane which are extracellular matrices holding cells and tissues together. They interact with receptors present in the plasma membrane of cells neighboring basement membranes. This interaction allows laminins to effectively regulate various cellular activities and signaling pathways.

Purified from EHS sarcoma according to the method of Timpl, et al.
Purified protein. Liquid; in 0.05 M Tris-HCl, pH 7.4, containing 0.15 M NaCl.

Preparation Note

Specificity: Molecular weight = 850 kD; 3 chains expressed as two major bands at 200 and 400 kD on SDS-PAGE.

Preparation: Prepared from Engelbreth-Holm-Swarm {EHS} sarcoma according to the method of Templ et al 1979 {http://www.jbc.org/cgi/reprint/254/19/9933.pdf} Briefly, tumor homogenates were subjected to NaCl extractions and centrifugations. Laminin was found in the 0.5M NaCl fraction. After subsequent salt gradients and further centrifugation steps, the laminin was dialyzed against 2M urea and passed over a column of DEAE, followed by molecular sieve chromatography and concentration by ultrafiltration.

Storage and Stability

Store at -20°C for up to six months.
Thaw product carefully at 2-8°C to avoid premature gelling, and do not refreeze. Once thawed. material should be stored at 2-8°C for up to 3 months.
