Description

General description

Nicotinamide adenine dinucleotide phosphate (NADP) and NADPH form a redox pair. The NADPH/NADP ratio regulates the intracellular redox potential, thereby influencing metabolic reactions in vivo.

Application

The β-NADH phosphate disodium salt has been used for the quantification of glucose in glycogen. It has also been used for the hexokinase assay.

Preparation Note

Storage conditions (working solution): 2 to 8 °C
10 mg/ml in aqueous solution, contains at least 99% NADP after 4 weeks at 2 to 8 °C

Analysis Note

Absorbance coefficients: 18.0 x 10³ [l x mol^-1 x cm^-1] at 260 nm, 5.9 x 10³ [l x mol^-1 x cm^-1] at 333 nm (CN complex)

Absorbance at 340 nm: Imidazol buffer, 100 mM pH 6.7, NADP 2 mM (= 14.87 mg/10 ml) : A₃₄₀ approximately 0.100.

Absorbance at 360 nm: Aqueous solution of NADP, 10 mg/ml: A₃₆₀ approximately 0.200-0.250.
