Human fibrosarcoma

Cell Line Description

Established from a fibrosarcoma arising adjacent to the acetabulum of a 35 year old caucasian male. The cells can give rise to tumours in immunosupressed NIH Swiss mice. Susceptible to infection by RNA tumour viruses including RD114 feline endogenous virus and FeLV.

Application

HT 1080 has been used to demonstrate that type I collagen induces matrix metallopeptidase 2 (MMP-2) activation. It has also been used to study the roles of Ras-related C3 botulinum toxin substrate 1 and 2 (Rac1 and Rac2) in regulating morphology, F-actin organization, adhesion, migration and chemotaxis of HT1080 cells.

Culture Medium

EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:4 to 1:8 i.e. seeding at 1-4x10,000 cells/cm2 using 0.25% trypsin/EDTA; 5% CO2; 37°C.
