General description

Monoclonal Anti-Sirt1 (Sir2) (mouse IgG1 isotype) is derived from the hybridoma SIR11 produced by the fusion of mouse myeloma cells (NS1 cells) and splenocytes from BALB/c mice immunized with a synthetic peptide corresponding to amino acids at the C-terminus of mouse Sir2 with C-terminal added lysine. Sirt proteins are a family of proteins that are found in organisms ranging from bacteria to complex eukaryotes. Members of this family contain a 250 amino acid core domain that shares about 25-60% sequence identity. Sirtuin 1 (SIRT1) protein, a mammalian homolog of yeast Sir2, is a NAD-dependent histone deacetylase.

Specificity

Monoclonal Anti-Sirt1 (Sir2α) antibody is specific for Sir2α in mice.

Monoclonal Anti-Sirt1 antibody is suitable for use in western blot (1-2 μg/mL using total cell extract of C-2 cells), immunoblot (approx. 105 kDa), microarray and indirect ELISA.

Biochem/physiol Actions

SIRT1 is a highly conserved sirtuin family deactelyase that modulates heart metabolism . SIRT1 has also been implicated in senescence and aging . Thus, SIRT1 may be used as a target for treating age-related diseases.
Sirt1 transfers acetyl groups from its protein substrates to ADP-ribose and synthesizes O-acetyl-ADP-ribose. Through histone deacetylation, Sirt1 may silence chromatin. It appears that Sirt1 NAD requirement makes this protein an important player in the pathway that leads to increased life span of several species through calorie restriction. Sirtuin 1 (SIRT1) plays an important role in neuronal proliferation and vascular cells. It also plays an athero-protective role. Mutations in the SIRT1 gene affects the metabolism, cholesterol and fat deposition. SIRT1 has the ability to intermediate the insulin resistance through an inflammatory process, reactive oxygen species, gluconeogenesis and adiponectin level.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
