General description

Macrophage colony stimulating factor 1 (CSF1,CSF-1, M-CSF, MCSF) or Lanimostim is a cytokine that was first identified as a molecule that plays an important role in the survival, proliferation and differentiation of mononuclear cells such as phagocytes, macrophages and monocytes. Today however we know that CSF1 also serves several other functions in a variety of cell types. CSF1 is essential for normal bone development, osteoclast proliferation, and the resorption of bone. CSF1 is released by osteoclasts upon stimulation by parathyroid hormone. Additionally CSF1 impacts actin and cytoskeleton reorganization which effects cell adhesion and migration, and overstimulation of CSF1 in endothelial cells is linked to the development and progression of atherosclerosis. CSF1 is also highly expressed in the endometrial epithelium of the pregnant uterus and CSF1 is essential for normal embryonic implantation and placental development. CSF1 appears to play an important role in cancer as well; first CSF1 is an anti-tumor agent and increases macrophage expression of anti-tumor activity, but over expression of CSF1 in breast, bone, and several other forms of cancer appears to promote cell invasiveness, proliferation, and tumor survival. Finally, CSF1 as a secreted mature factor promotes the release of proinflammatory cytokines and serves as part of the innate immunity process for inflammation and infection. CSF1 is initially produced as a precursor that is membrane bound but processed and secreted upon stimulation. CSF1 is heavily glycosylated and found both in the ECM as well as surface membranes of cells. CSF1 receptor is found on numerous cell types including macrophages, endothelial cells, and osteoclasts. EMD-Millipore’s Anti-CSF1 mouse monoclonal antibody has been tested in western blot on recombinant protein and in paraffin embedded immunohistochemistry on human placental tissue sections.

Application

Anti-CSF1 Antibody, clone 2D10 is a highly specific mouse monoclonal antibody, that targets CSF1 & has been tested in western blotting & IHC.
Immunohistochemistry Analysis: A 1:200-1,000 dilution from a representative lot detected CSF1 in human placenta tissue.

Optimal working dilutions must be determined by end user.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
