General description

The gene zinc transporter-4 (ZNT4) is mapped to human chromosome 15q21.1. ZNT4 belongs to family of zinc transporters (ZNTs). ZNTs have six transmembrane domains and histidine rich loops which connect these domains. ZNT4 is ubiquitously expressed and is localized in the plasma membrane and lysosomes.

Application

Biochem/physiol Actions

Zinc transporter-4 (ZNT4) transports Zn from the cytosol to lysosomes and endosomes. siRNA-mediated down-regulation of ZNT4 eliminates the Zn-induced lysosomal swelling and Zn retention in TRPML1 (mucolipin-1)-deficient HeLa cells. Premature termination codon in ZNT4 decreases the transport of Zn into milk in mouse model. ZNT4 is involved in the pathological process which causes β-amyloid peptide containing senile plaque formation in Alzheimer′s disease. ZNT4 is down-regulated in prostate benign prostatic hyperplasis and tumor samples.
