General description

The gene PREB (prolactin regulatory element binding) is also referred to as Sec12 and is mapped to human chromosome 2p23. The encoded 417 amino acids protein contains highly conserved seven WD-40 folds. It is localized to the ER (endoplasmic reticulum) exit sites in mammalian cells. The encoded protein contains two PQ-rich potential transactivation domains and lacks a DNA binding motif. This gene overlaps the αβhydrolase domain containing 1 (ABHD1) gene along a stretch of 42bp and may be associated with some of the developmental abnormalities found in patients with partial trisomy 2p.

Application

Biochem/physiol Actions

The gene PREB (prolactin regulatory element binding) encodes a type II transmembrane protein. It binds to the Pit1-binding element of the prolactin (PRL) promoter and plays a transcriptional regulatory role in the pituitary and other organs in which the protein is expressed. In response to cytokines, PREB transcriptionally regulates MCP-1 (monocyte chemoattractant protein-1) promoter. MCP-1 functions as an initiating factor in the inflammatory process of atherogenesis. PREB also functions as a guanine-nucleotide exchange factor for Sar1 guanosine triphosphatase and is essential for collagen VII secretion. It is recruited to ER (endoplasmic reticulum) exit sites by its interaction with a collagen cargo receptor component, cTAGE5. The export of large cargo at the ER exit sites involves high levels of guanosine triphosphate-bound Sar1 generated by PREB.
