Paired box protein Pax-1 (UniProt P15863; also known as HuP48) is encoded by the PAX1 (also known as HUP48, OFC2) gene (Gene ID 5075) in human. Pax-1 belongs to the family of transcription factors with conserved DNA-binding paired box domain. The family of paired box (PAX) genes are divided into subgroup I (PAX1 and PAX9), II (PAX2, PAX5, and PAX8), III (PAX3 and PAX7), and IV (PAX4 and PAX6). Pax1 & Pax9 are paralogous genes with overlapping expression and function. Pax1 is required for the proper development of the ventral structures of the vertebral column and double homozygous mutants of Pax1 and Pax9 result in a dramatic increase in the severity of the vertebral defects. During the early stages of chondrogenic differentiation, Pax1 induces and transactivates the transcriptional repressor Nk3 homeobox 2 (Nkx3.2) that promotes chondrogenesis of sclerotomal cells, while Pax1 act as a negative regulator during chondrogenic differentiation by antagonizing against Sox9-driven chondrocyte maturation. Pax1 is downregulated as cartilage formation proceeds and becomes restricted to the mesenchymal condensations that give rise to the anlagen of the intervertebral discs, and to the perichondrium surrounding the cartilaginous vertebral bodies.
Specificity
Reactivity toward spliced isoforms 2 and 3 has not been determined.
