General description

Penicillinase from Bacillus cereus has been shown to contain three distinct species when purified and examined for homogeneity by sedimentation analysis, amino-terminal analysis, and vertical acrylamide gel electrophoresis. [1]

Application

Used in the production of penicillin.[2] Penicillinase from Bacillus cereus has been used in a comparison study with penicillinase from Bacillus subtillis where the Michaelis constant, shape of the Ph/activity curve and immunological properties were shown to be significantly different.
Penicillinase can be used to reduce levels of penicillin in patients with severe penicillin reactions. [3]
Used in the production of penicillin.

Biochem/physiol Actions

Penicillinase specifically catalyzes the hydrolysis of β-lactam ring of penicillin.

Protein determined by biuret.

Other Notes

One unit will hydrolyze 1.0 μmole of indicated substrate per min at pH 7.0 at 25 °C. Sold as benzylpenicillin units. This International Unit (using benzylpenicillin as substrate) is approximately equal to 600 Levy or 75 Pollock units.

  2. I have just received the product P0389-1KU 1000 units. The protocol I'm going to use requires me to use my 10 mg/ml solution. However, I don't understand what your "1000 Units" of product correspond to. Could you please give me more information pls?

    1. The amount of powder in the vial will depend on the activity reported on the lot specific Certificate of Analysis. For example, lot 0000211383 is reported to have an activity of 115 units per milligram. A package of 1000 units would contain 8.7 mg. The 1000 unit package would prepare 870ul of a 10 mg/mL solution.
