General description

Platelet-Derived Growth Factor (PDGF), a cationic polypeptide, is an important serum mitogen predominantly found in cells of mesenchymal origin. PDGF is mapped to human chromosome 7p22.3 and is expressed in smooth muscle cells, endothelial cells and macrophages. PDGF exists as hetero and homodimers : PDGF-AB FDGF-AA, PDGF-BB, PDGF-CC and PDGF-DD.

Application

Platelet-Derived Growth Factor-BB human has been used in induction of DNA synthesis and proliferation in low-concentration serum.

Biochem/physiol Actions

Platelet-derived growth factor (PDGF) is a potent chemoattractant and activator of neutrophils, monocytes, and fibroblasts. PDGF elicits multifunctional actions in a variety of cells. It is mitogenic to mesoderm-derived cells, such as dermal and tendon fibroblasts, vascular smooth muscle cells, glial cells and chondrocytes. PDGF-BB isomer is highly expressed in tumor, mediating oncogenic pericyte recruitment and is is a key regulator of angiogenesis. PDGF-BB controls bone homeostasis, repair and regeneration. It increases the synthesis of phospholipids, cholesterol esters, glycogen and prostaglandins and modulates LDL receptor binding.

The biological activity is measured by the dose-dependent stimulation of thymidine uptake in BALB/c 3T3 cells.
