General description

A synthetic cyclic alkylamino luciferin that exhibitsproperties that are superior to those of D-luciferin. Binds to luciferase with higheraffinity and emits about 5.7-fold more light than aminoluciferin and3.2-fold more light than D-luciferin. Its superior light output is maintainedover a wide range (1-100 µM). Shown to readily cross the blood-brain barrier inmice and provides much improved noninvasive bioluminescence imaging (BLI) ofthe brain at a lower dose when compared to D-luciferin. Yields about 10-foldhigher bioluminescent signal than D-luciferin when Luc2 luciferase expressing4T1 breast cancer cells are implanted into the mammary fat pads of BABL/c mice.A rapid (4-5 min), long-lasting, and steady signal peak is observed followingan i.v injection of CycLuc1 in mice.

Application

Luciferase Substrate, CycLuc1 has been used for transfecting HEK293 cells to study the inhibition of intracellular luciferase crystallization by D-luciferin and its synthetic analogues.
