General description

Protein tyrosine phosphatase, receptor type S (PTPRS) also known as PTPσ belongs to the subfamily of receptor-type protein tyrosine phosphatases. It is encoded by the gene mapped to human chromosome 19p13.3. PTPRS consists of large ectodomain and a membrane-spanning intracellular domain.
Human PTPRS (GenBank Accession No. BC104812) (amino acids 883-end), with N-terminal GST tag, MW = 98kDa, expressed in a Escherichia coli expression system.

Application

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.

Biochem/physiol Actions

Protein tyrosine phosphatase, receptor type S (PTPRS)/ PTPs localized on the cell surface plays a vital role in cell–cell and cell– extracellular matrix (ECM) interactions, and is implicated in the regulation of axon homeostasis and neuronal development. In addition, PTPRS also has a crucial role in autophagy regulation. PTPRS expressed on plasmacytoid dendritic cells (pDCs) inhibits the production of type I interferon (IFN) in response to viruses. Mutation of both Ptprs and Ptprf (protein tyrosine phosphatase, receptor type F) results in pDC hyperactivation and mild colitis.

Unit Definition

One unit will hydrolyze 1 nmol p-nitrophenyl phosphate per minute at pH 7.4 and 30°C.

Physical form

Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 20% glycerol and 3 mM DTT.

Preparation Note

Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.
