Description générale

Biotherapeutics, for example, bio-engineered drugs and peptide therapeutics, represent the promise of new medical treatments for the future. Production costs have been falling leading to extremely high demand for suitable analytical methods for process monitoring and quality control of these biomolecules. HPLC is the preferred method of analysis, and it is therefore important to use the right column for these larger molecules.Accurate analysis of proteins, antibodies, and large peptides requires columns with good permeability, along with better mass transfer and selectivity. In order for size-exclusion not to influence the separation, the pore size should be approximately ten times larger than the molecule being analyzed.Chromolith® WP 300 columns are made of monolithic silica with a characteristic bimodal pore structure.Chromolith® WP 300 RP-4 columns have macropores with 2 µm diameter, giving a column efficiency exceeding 80,000 plates/meter. The mesopores are 30 nm (300 Å), the surface modification is Butylsilane. Chromolith® WP 300 RP-4 columns fall under the USP L26 Classification. The column efficiency is comparable to a 5 µm particle size fully porous particulate column. Chromolith® WP 300 RP-4 columns are suitable for LC-MS applications.The column dimension of 50 mm length and 4.6 mm I.D. is the best choice for very fast separations of Biomolecules on any HPLC System. Chromolith® WP 300 RP-4 50-4.6 HPLC columns enable:

  • High-speed separations at very low column back-pressure
  • High matrix tolerance, allowing time and cost savings with simplified workflows
  • Cost savings due to extended column lifetimes
  • Connection of columns, flow gradients, and the use of mobile phases with higher viscosity
The recommended Guard-columns are: Chromolith® WP 300 RP-4 5-4.6 guard cartridges (3 pieces) [1.52263] with the Chromolith® Guard Cartridge Holder Bioinert 5 mm x 4.6 mm [1.52255] Or Chromolith® WP 300 RP-4 10-4.6 guard cartridges (3 pieces) [152262] with the Chromolith® Guard Cartridge Holder Bioinert 10 mm x 4.6 mm [1.52256] The Chromolith® column coupler can be used to significantly increase the separation efficiency of Chromolith® columns by coupling several columns in series.
