Biochem/physiol Actions

WIC1 is a potent Wnt/β-catenin signaling inhibitor (IC50 = 10 nM against 24h 5 μM CHIR-induced BEAS2B TCF/LEF reporter activity) that reduces airway basal stem cell (ABSC) proliferation (92% reduction of 4d 2 μM CHIR-induced hABSC proliferation by 1 μM WIC1) and induces ciliated cell differentiation (by 1.9-fold on d14 with 100 nM WIC1; mABSCs under air-liquid interface (ALI) condition) by suppressing β-catenin Y489 phosphorylation (complete inhibition of 24h 5 μM CHIR-induced pY489 in BEAS2B by 1 μM WIC1) and TP63 expression (80% reduction of 9d 1 μM CHIR-induced TP63 in mABSC by 1 μM WIC1; 40% reduction of 48h 5 μM CHIR-induced TP63 in BEAS2B by 1 μM WIC1).
