General description

Enrichment of T7 phage displaying target peptides or proteins can easily be accomplished with the T7Select® Biopanning Kit. The kit contains reagents to perform biopanning with target ligands using 96-well microtiter plates or similar solid phases. Biopanning procedures can be tested and optimized using the positive and negative controls provided in the kit. The supplied Positive Control Lysate contains a T7Select415-1 recombinant that displays the S•Tag peptide. When combined with the Negative Control Lysate at known ratios, the degree of enrichment of high affinity binding phage can be monitored by a simple plaque lift assay with the provided detection reagents.
Biopanning selection
Biopanning requires both binding and elution conditions that maintain the infectivity of the phage. The stability of the T7 phage particle enables the use of a variety of elution conditions during biopanning. Phage maintain infectivity following treatment with 1% SDS, 5 M NaCl, 4 M urea, 2 M guanidine-HCl, 10 mM EDTA, reducing conditions (up to 100 mM DTT), and a pH range of 4 to 10. The wide range of conditions available for T7Select biopanning expands the range of targets that can be used successfully.
