Specificity

Clone 1B9 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Ubiquitin-protein ligase E3A (UBE3A). It targets an epitope within 23 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the N-terminal half of human Ubiquitin-protein ligase E3A (UBE3A).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected UBE3A in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected UBE3A in U251-MG cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected UBE3A in Human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound UBE3A peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected UBE3A in HeLa cells.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected UBE3A in one million HeLa cells.

Target description

Ubiquitin-protein ligase E3A (UniProt: Q05086; also known as EC: 2.3.2.26, E6AP ubiquitin-protein ligase, HECT-type ubiquitin transferase E3A, Human papillomavirus E6-associated protein, Oncogenic protein-associated protein E6-AP, Renal carcinoma antigen NY-REN-54) is encoded by the UBE3A (also known as E6AP, EPVE6AP, HPVE6A) gene (Gene ID: 7337)in human. UBE3A is the E3 ubiquitin-protein ligase that accepts ubiquitin from an E2 ubiquitin-conjugating enzyme in the form of a thioester and transfers it to its substrates. Several substrates have been identified, including the RAD23A and RAD23B, MCM7, annexin A1, the PML tumor suppressor, and the cell cycle regulator CDKN1B. UBE3A catalyzes the high-risk human papilloma virus (HPV) E6-mediated ubiquitination of p53/TP53, contributing to the neoplastic progression of cells infected by these viruses. Phosphorylation at Tyr659 by ABL1 impairs E3 ligase activity and protects p53/TP53 from degradation in (HPV)-infected cells. UBE3A may function as a cellular quality control ubiquitin ligase by helping the degradation of the cytoplasmic misfolded proteins and it also promotes its own degradation in vivo. It is reported to plays an important role in the regulation of the circadian clock by ubiquitinating the core clock component ARNTL/BMAL1 and leading to its proteasomal degradation. Three isoforms of UBE3A have been reported that are generated via alternative splicing. Mutations in UBE3A gene cause Angelman syndrome that is characterized by severe motor and intellectual retardation, ataxia, hypotonia, and frequent jerky limb movements. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
