Specificity

Clone IF21 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects IL-17A. It targets an epitope within 21 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the internal region of human Interleukin-17A (IL-17A).

Application

Quality Control Testing

Evaluated by Western Blotting in Mouse Thymus tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected IL-17A in Mouse Thymus tissue lysate.

Tested applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected IL-17A in HL-60 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected IL-17A in human colon and human thymus tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound IL-17A with a KD of 3.4 x 10-8 in an affinity binding assay.

Flow Cytometry Analysis: 1 μg from a representative lot detected IL-17A in one million HL-60 cells.

Target description

Interleukin-17A (UniProt: Q16552; also known as IL-17, IL-17A, Cytotoxic T-lymphocyte-associated antigen 8, CTLA-8) is encoded by the IL17A (also known as CTLA8, IL17) gene (Gene ID: 3605) in human. IL-17A is a proinflammatory cytokine secreted by activated CD4+ Th17 cells. It is synthesized with a signal peptide (aa 1-23) that is subsequently cleaved off to produce the mature protein. Its levels are up-regulated by IL23A-IL12B. It is an effector cytokine of innate and adaptive immune system involved in antimicrobial host defense and maintenance of tissue integrity. It plays important roles in multiple biological processes, including the induction of IL-6, IL-8, G-CSF, and PGE2 production in epithelial, endothelial or fibroblasts; the enhancement of surface expression of ICAM-1 in fibroblasts; activation of NF-kB and co-stimulation of T cell proliferation. It is also involved in antimicrobial host defense and maintenance of tissue integrity. Its actions are mediated via interaction with IL-17RA and IL-17RC heterodimeric receptor complex, triggering homotypic interaction of these receptors with TRAF3 Interacting Protein 2 (TRAF3IP2) adapter. This leads to downstream TRAF6-mediated activation of NF-kB and MAP kinase pathways resulting in transcriptional activation of cytokines, chemokines, antimicrobial peptides, and matrix metalloproteinases. IL-17A also plays an important role in connecting T cell-mediated adaptive immunity and acute inflammatory response to destroy extracellular bacteria and fungi. In synergy with IL-17F, it is shown to mediate the production of antimicrobial b-defensins DEFB1, DEFB103A, and DEFB104A by mucosal epithelial cells, which limits the entry of microbes through the epithelial barriers. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
