General description

AATF/Rb-binding protein Che-1, also known as Protein Apoptosis-antagonizing transcription factor (Protein AATF), Rb-binding protein Che-1, and encoded by the gene name AATF/ CHE1/ DED/ HSPC277, is a member of the AATF family. Recent studies have indicated that AATF may function as a general inhibitor of the histone deacetylase HDAC1. AATF binds to the pocket region of RB1, and consequently it may displace HDAC1 from RB1/E2F complexes, leading to activation of E2F target genes and cell cycle progression. Conversely, displacement of HDAC1 from SP1 bound to the CDKN1A promoter leads to increased expression of this CDK inhibitor and blocks cell cycle progression. In addition, AATF antagonizes PAWR mediated induction of aberrant amyloid peptide production in Alzheimer disease (presenile and senile dementia), although the molecular basis for this phenomenon has not been described to date. AATF/Rb-binding protein Che-1 is expressed at high levels in brain, heart, kidney, placenta and thymus.

Quality

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:500 dilution of this antibody detected AATF/Rb-binding protein Che-1 in 10 µg of HeLa cell lysate.

Target description

~70 kDa observed. The calculated molecular weight is 63 kDa, however AATF/Rb-binding protein Che-1 has been shown as a ~70 kDa band in western blots (Guo, G., et al. (2004). J. Biol. Chem. 279:4596-4603).

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
