General description

An antibiotic that inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit, preventing binding of aminoacyl tRNA to the A-site and blocking translocation. Used commonly for selection of the bacterial tetr gene whose product, a 399-amino acid membrane-associated protein, functions in active export of the drug from the cell.

Reconstitution

Following reconstitution, sterilize by filtration through a 0.22 µm pore-size filter, aliquot and freeze (-20°C). Stock solutions are stable for 1 month at -20°C. Agar plates are stable for 2 weeks at 4°C. Autoclaved media should be cooled to ≤55°C before adding tetracycline.
