Alpha Synuclein

StressMarq’s alpha synuclein pre-formed fibrils (PFFs) seed the formation of new fibrils from active alpha synuclein monomers, and can be used to induce endogenous alpha synuclein phosphorylation and subsequent Lewy body inclusion formation in neuronal cell culture or for in vitro oligomerization studies. Alpha synuclein oligomers are increasingly thought to be the more toxic species in synucleinopathies. StressMarq’s kinetically stable alpha synuclein oligomers are toxic to dopaminergic neurons and induce phosphorylation of alpha synuclein Ser129 (a pathology associated with Parkinson’s disease) and are generated without the addition of any inducers or inhibitors. Alpha synuclein monomers are capable of aggregation and do not show neurodegenerative activity.

Tau

StressMarq’s active tau pre-formed fibrils (PFFs) and monomers are available to study tau aggregation, a hallmark of neurodegenerative diseases including Alzheimer’s. The process of tau aggregation can be seeded by active tau PFFs, which recruit monomers to form larger tau fibrils. This is demonstrated in thioflavin T assays where an increase in fluorescence (indicative of tau fibrillization) is seen when tau PFFs are combined with tau monomers. Tau monomers and PFFs are available as wild-type or with a variety of mutations. Several tau constructs are available expressed in Baculovirus in addition to E. coli. Baculovirus tau PFFs are post-translationally modified and fibrillize without a scaffold.

Amyloid Beta

StressMarq’s amyloid beta pre-formed fibrils (PFFs) present as long strands when observed under TEM and AFM and have a unique high molecular weight signal on a Western Blot with an amyloid beta antibody. Our amyloid beta PFFs also demonstrated to be toxic to primary rat cortical neurons in a dose-dependent manner after an initial sonication step. Our amyloid beta oligomers present as globular oligomers when observed under TEM and AFM and have a unique dimer/trimer and oligomer signal on a Western Blot. In the brain, amyloid beta peptide is generated by protease cleavage of amyloid precursor protein (APP), which aggregates into oligomers, protofibrils, fibrils and ultimately plaques in neurodegenerative diseases.
