General description

Tris Buffered Immunohistochemistry wash buffer plus TWEEN 20 is a 20X concentrated solution that is employed to rinse reagents off slides and to provide a medium for short-term storage of immunohistochemistry specimens in between application of reagents.

Biochem/physiol Actions

Tween 20 helps in the enhancement of intestinal movement of doxorubicin in vitro but not in vivo.

  1. Is there a recommended or preferable wash buffer (TBS or PBS) for a breast panel in Autostainer? Moreover, please confirm how long one can wait once the dilution of the concentrated product is done? Both TBS and PBS in this case.

    1. Phosphate Buffer Saline (PBS) and Tris Buffer Saline (TBS) are concentrated wash solutions designed for rinsing reagents from tissue slides and preserving tissue sections between reagent applications. PBS is recommended with any detection system, except those containing alkaline phosphatase. Conversely, TBS is recommended when alkaline phosphatase is present to avoid excessive phosphate accumulation and consequent reduction in the rate of reaction. Adding Tween® 20 to both PBS and TBS may help reduce background and produce a cleaner immunohistochemical stain overall. The most significant factor is the intention for use to visualize the stain, with what chromogen and detection kit. For HRP or Alk Phos on the breast panel, TBS can be used for both. For diluent, as long as products are stored properly (2 to 8C), the diluent will help stabilize diluted antibodies, and the same applies for use with HRP or Alk Phos detection for TBS to be run on either, while PBS works best with HRP. This allows for a longer expiration date and aligns with the antibody expiration date.
