Specificity

Clone 2G22 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Syk. It targets an epitope within 16 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the C-terminal half of human Tyrosine-protein kinase SYK (Syk).

Application

Quality Control Testing

Evaluated by Western Blotting in Ramos cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Syk in Ramos cell lysate.

Tested applications


Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Syk in lysates from A431 and Raw264.7 cells and Mouse Spleen tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Syk in human tonsil tissue sections.

Flow Cytometry Analysis: 1 μg from a representative lot detected Syk in one million Ramos cells.

Affinity Binding Assay: A representative lot of this antibody bound Syk with a KD of 6.6 x 10-8 in an affinity binding assay.

Target description

Tyrosine-protein kinase SYK (UniProt: P43405; also known as EC:2.7.10.2, Spleen tyrosine kinase, p72-Syk) is encoded by the SYK gene (Gene ID: 6850) in human. Syk is a non-receptor tyrosine kinase that is highly expressed in hematopoietic cells. It plays a crucial role not only in adaptive immune receptor signaling, but also in cellular adhesion, innate immune recognition, osteoclast maturation, platelet activation and vascular development. Syk contains two SH2 domains (aa 15-107 and 168-259) that mediate its interaction with the phosphorylated ITAM domains of transmembrane proteins. Its protein kinase domain is localized in amino acids 371-631. Intramolecular binding of the interdomains A and B (aa 108-160 and 260-370) to parts of its catalytic domain keeps the catalytic center in an inactive conformation. The phosphorylation of the interdomains or the binding of the SH2 domains with dually phosphorylated ITAM domains on transmembrane proteins disrupts these intramolecular interactions and allows the kinase domain to adopt an active conformation. Initial ITAM binding triggers rapid activation of Syk, whereas subsequent phosphorylation leads to prolonged activation. The phosphorylation of Syk and of the ITAM domains is essentially mediated by SRC subfamily kinases, like LYN, upon transmembrane receptors engagement. Syk can phosphorylate ITAMs and autophosphorylate its own interdomain region tyrosines, providing a positive feedback loop following initial ITAM-mediated Syk activation. Syk is also reported to mediate signaling of receptors that do not contain an ITAM, including integrins and C-type lectins, via interaction with ITAM-containing adaptors, such as DAP12 (TYROBP) and the FcR -chain (FcR ). This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
