General description

CF647 goat anti—human IgM (μ chain) is labeled with our bright, deep red-fluorescent CF647 dye and is prepared from antibodies purified by human IgM—affinity chromatography. The antibodies react specifically with IgM heavy chains (μ chains) and not with immunoglobulin light chains. To minimize cross-reactivity, it has been adsorbed human IgG prior to conjugation
During the B-cell development, IgM (immunoglobulin M) is the first expressed antibody present as a monomer on the surface of naive B cells. Antigenic stimulation and upon maturation, the monomeric IgM units (190kDa), join together to form a pentameric immunoglobulin structure. It also but rarely forms a hexameric structure. The pentameric IgM is linked by a disulfide bond, a polypeptide chain and a J-chain. The μ-chain consists of five domains- VH, Cμ1, Cμ2, Cμ3 and Cμ4.

Specificity

Binds human IgM heavy chain (mu chain) (minimal cross-reactivity IgG and IgA heavy chains, or immuglobulin light chains).

Biochem/physiol Actions

The J-chain of the multimeric IgM (immunoglobulin M) structure is responsible for secretion at the mucosal surfaces. The pentameric form of IgM, has a greater affinity towards antigens (with multiple repeating epitopes) compared to the monomers. The pentameric IgM enables better opsonization of antigens for destruction and fixing complement. IgM is essential of the activation of classical complement system. Reduced serum IgM levels is observed in Bruton′s agammaglobulinemia an X-linked disorder and transient hypogammaglobulinemia in infants.

Physical form

Supplied in phosphate buffered saline with 0.05% sodium azide, 50% glycerol and 0.2% BSA.

Preparation Note

Protect from light. The antibody solution should be gently mixed before use.
