General description

Podocalyxin-like protein 1 (PODXL) is a transmembrane protein known to be heavily sialyated and sulfated that is localized on the apical surface of podocytes and on vascular endothelia. Podocalyxin protein is expressed on the surface of undifferentiated human embryonic stem cells. Originally, the protein was characterized as a marker for vasculature and podocytes, but expression has since been observed in platelets, hematopoietic precursor cells and mesothelial cells from the coelmic cavity lining. Studies have outlined two distinct and seemingly adverse roles for this protein; it acts as an anti-adhesion molecule in renal glomerulae podocytes, but functions as an adhesion molecule within vasculature on high endothelial venules. Podocalyxin expression has been implicated in the development of aggressive cancers, beta catenin signaling, angiogenisis, and both myeloid and acute lymphoblastic leukemias.

Specificity

This antibody recognizes the extracellular domain of Podocalyxin-like protein 1.
This antibody has been shown to select against undifferentiated human embryonic stem cells (hESC) and to be functionally cytotoxic against these undifferentiated hESC populations (Choo, A., et al., 2008).

Application

Anti-Podocalyxin-like protein I (Cytotoxic) Antibody, clone mAb 84 is an antibody against Podocalyxin-like protein I (Cytotoxic) for use in Western Blotting, ICC, Flow Cytometry.
This antibody was shown to induce cell death of undifferentiated, but not differentiated hESC, within 30 minutes of incubation (Choo, A., et al. (2008). Stem Cells 26:1454-1463). This antibody has been shown to work in Western Blot applications (Choo, A., et al. (2008). Stem Cells, 26:1454–1463).
This antibody has been shown to work in Flow Cytometry applications (Choo, AB., et al. (2008). Stem Cells, 26:1454–1463).

Quality

Evaluated by Immunocytochemistry in H9 human embryonic stem cells. Immunocytochemistry Analysis: A 1:500 dilution of this antibody detected Podocalyxin-like protein I in H9 human embryonic stem cells.

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
