General description

24-hydroxycholesterol (24-OH) is an oxidized product of cholesterol. It is synthesized in the presence of enzyme 24S-hydroxylase (CYP46A1).

Biochem/physiol Actions

24(S)-hydroxycholesterol is preferred over 24(R)-hydroxycholesterol by human 7α-hydroxylase. However, the porcine enzyme is active towards both isomers. 24-hydroxycholesterol (24-OH) favors sirtuin 1 expression and reduces the tau protein phosphorylation.

(24S)-Hydroxycholesterol is formed from cholesterol in the brain and is important for cholesterol homeostasis in this organ. Elimination of (24S)-hydroxycholesterol has been suggested to occur in the liver but little is known about the metabolism of this oxysterol. In the

It is now established that cholesterol oxidation products (oxysterols) are involved in several events underlying Alzheimer's disease (AD) pathogenesis. Of note, certain oxysterols cause neuron dysfunction and degeneration but, recently, some of them have been shown also to have neuroprotective
