General description

Kaposi′s sarcoma-associated herpesvirus glycoprotein K8.1 (UniProt: O36551; also known as Glycoprotein K8.1, Glycoprotein K8.1A, Glycoprotein gp35/37, K8.1, K8.1 protein) is encoded by the K8.1 (also known as HHV8GK18_gp55) gene (Gene ID: 4961469) in Kaposi′s sarcoma-associated herpesvirus (KSHV). Kaposi sarcoma virus is an oncogenic human g-herpesvirus that has been identified in AIDS patients in association with Kaposi′s sarcoma. It can persist latently in host cells while expressing only a limited set of viral genes and maintaining its genome as extrachromosomal plasmids. This virus encodes two immunogenic class I glycoproteins, K8.1A and K8.1B. These glycoproteins have several serine and threonine residues that serves as potential sites for O-glycosylation. The 228-amino-acid-long K8.1A open reading frame (ORF) contains four N-glycosylation sites and the 167-amino-acid-long K8.1B ORF contains three N-glycosylation sites. Glycoprotein K8.1A is reported to mediate KSHV binding to target cells via interaction with heparan sulfate. This protein is involved in binding to target cells but is not required for viral entry into cells. Clone 4A4 detects both K8.1A and K8.1B glycoprotein and is shown to react with infected cell membranes and virion envelope. This antibody is shown to partially inhibit KSHV infection in HEK293 cells. Specificity

Clone 4A4 is a mouse monoclonal antibody that detects Glycoprotein K8.1 of Kaposi′s sarcoma-associated herpesvirus.

Evaluated by Western Blotting in Phorbol-12-myristate-13-acetate (TPA)-induced BCBL-1 cells.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected Kaposi′s sarcoma-associated herpesvirus (KSHV) in Phorbol-12-myristate-13-acetate (TPA)- induced BCBL-1 cells.

Tested Applications

Inhibition: A representative lot of this antibody inhibited infection by KSHV in HEK293 cells.(Faure, A., et al. (2019). Proc Natl Acad Sci USA. 116(33):16519-16528).

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
