General description

Transformation/transcription domain-associated protein (UniProt Q9Y4A5; also known as 350/400 kDa PCAF-associated factor, PAF350/400, STAF40, TR-AP, Tra1 homolog) is encoded by the TRRAP (also known as PAF400) gene (Gene ID 8295) in human. Transformation/transcription domain-associated protein (TRRAP) is a cofactor component shared by several histone acetyltransferase (HAT) complexes, including SAGA (Spt-Ada-Gcn5-acetyltransferase), PCAF (P300/CBP-associated factor), and Tip60. TRRAP is an essential component of the histone epigenetic modifying mechinery that plays an important role in regulating cell-cycle progression. Trrap gene-knockdown or protein downregulation causes cell and mouse lethality, impaired spindle checkpoint and differentiation.

Specificity

The immunogen sequence is present in both spliced isoforms of human TRRAP reported by UniProt (Q9Y4A5).

Epitope: N-terminal to nuclear localization signal in the TP53-interacting region.
Ovalbumin-conjugated linear peptide corresponding to a sequence from the TP53-interacting region of human TRRAP.

Application

Anti-TRRAP Antibody, clone 2TRR-1B3 is an antibody against TRRAP for use in Western Blotting, Immunoprecipitation.
Immunoprecipitation Analysis: 10 µL from a representative lot immunoprecipitated TRRAP from 1 mL (4.5 mg/mL) of HeLa nuclear extract (Courtesy of Dr. Mustapha OULAD-ABDELGHANI, IGBMC, France).
Immunoprecipitation Analysis: A representative lot immunodepleted TRRAP using nonhomologous end-joining/NHEJ-competent protein extracts prepared from human AAH-1 lymphoblastoid cells (Robert, F., et al. (2006). Mol. Cell. Biol. 26(2):402-412).

Quality

Evalulated by Western Blotting in HeLa nuclear extract.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TRRAP in 10 µg of HeLa nuclear extract.

Target description

~400 kDa observed. 437.6 kDa (isoform 1) and 434.4 kDa (isoform 2) calculated. Uncharacterized band(s) may appear in some lysates.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
