Cell Line Origin

Monkey African green kidney, SV40 transformed, serum-free

Cell Line Description

COS-7 cell line (Sigma catalog no. 87021302) adapted to grow in the chemically defined protein- and peptide-free InVitrus medium (Cell Culture Technologies). This cell line is substrate dependent, i.e., adherent. COS-7 was derived from CV-1 simian cells transformed by an origin-defective mutant of SV40.

Culture Medium

InVitrus medium (Cell Culture Technologies http://www.cellculture.com/) + 4 mM L-Glutamine (G7513) + 5 ug/ml Insulin. When freezing cells down use culture medium (50:50 conditioned medium:fresh medium) + 10% DMSO (D2650).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:2, i.e., seeding at 5x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37 °C. Please note: As the cell culture medium does not contain protein it will not inactivate trypsin. Once cells have detached from the flask it is necessary to dilute the trypsin with the cell culture medium, centrifuge the cells and resuspend in fresh medium. When freezing cells down use 50:50 fresh culture medium:conditioned medium plus 10% DMSO.
