Specificity
Clone 1B6 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Serum paraoxonase/arylesterase 2 (PON2). It targets an epitope within 17 amino acids from the N-terminal half. Predicted to react with all three isoforms.

KLH-conjugated linear peptide corresponding to 17 amino acids from the N-terminal half of human serum paraoxonase/arylesterase 2 (PON2).
Application
Quality Control Testing
Evaluated by Western Blotting in U87-MG cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PON2 in U87-MG cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PON2 in HeLa cell lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PON2 in Human liver tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound PON2 peptide with a KD of 3.5 x 10-8 in an affinity binding assay.

Target description
Serum paraoxonase/arylesterase 2 (UniProt: Q15165; also known as EC:3.1.1.2, PON 2, Aromatic esterase 2, A-esterase 2, Serum aryldialkylphosphatase 2) is encoded by the PON2 gene (Gene ID: 5445) in human. The paraoxonase (PON) gene family in humans consists of three conserved members, PON1, PON2, and PON3. They exhibit high similarity in their structural characteristics and have about ∼65% identity in their amino acids. PON2 is a membrane and peripheral membrane associated enzyme that is widely expressed with highest levels in liver, lung, placenta, testis, and heart tissue. It is capable of hydrolyzing lactones and a number of aromatic carboxylic acid esters and exhibits antioxidant activity. It serves as an intracellular guardian against oxidative stress. It is shown to prevent LDL lipid peroxidation and reverse the oxidation of mildly oxidized LDL (MM-LDL). It inhibits the ability of MM-LDL to induce monocyte chemotaxis. PONS2 overexpressing cells exhibit significantly less intracellular oxidative stress following treatment with hydrogen peroxide or oxidized phospholipid. These cells are also less effective in oxidizing and modifying LDL. PON2 is also involved in modulation of ER stress and regulation of cell proliferation/apoptosis. During ER stress, PON2 reduces the induction of proapoptotic CHOP protein, particularly the protein kinase JNK pathway, and also protects intrinsic apoptotic signaling, by prevention of mitochondrial superoxide formation and caspase activation. Three isoforms of PON2 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity™, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
