Specificity

Clone 1M6 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects S100-A9. It targets an epitope within 24 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal half of human S100-A9/MRP14.

Application

Quality Control Testing

Evaluated by Western Blotting in human tonsil tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected S100-A9/MRP14 in human tonsil tissue lysate.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound S100-A9/MRP14 peptide with at least one thousand-fold (1,000X) higher than with non-specific control peptide.

Immunofluorescence Analysis: A 1:1,000 dilution from a representative lot detected S100-A9/MRP14 in human tonsil tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected S100-A9/MRP14 in human tonsil tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Protein S100-A9 (UniProt P06702; also known as Calgranulin-B, Calprotectin L1H subunit, Leukocyte L1 complex heavy chain, Migration inhibitory factor-related protein 14, MRP-14, p14, S100 calcium-binding protein A9, S100A9) is encoded by the S100A9 (also known as CAGB, CFAG, CGLB, L1AG, LIAG, MAC387, MRP14) gene (Gene ID 6280) in human. S100A9 is one of the 21 members of the S100 family of proteins. S100A8 (calgranulin-A), S100A9 (calgranulin-B) and the heterodimer S100A8/S100A9 (calprotectin) exert their immunological and inflammatory action via binding to TLR-4, RAGE and some other putative receptors. Predominantly, the protein is localized in the cytoplasm and with increased intracellular calcium levels it is translocated from the cytoplasm to the cytoskeleton and the cell membrane. Calprotectin (S100A8/9) is predominantly expressed in myeloid cells with exception in inflammatory conditions where the expression is restricted to a specific stage of myeloid differentiation and both proteins are expressed in circulating neutrophils and monocytes but are absent in normal tissue macrophages and lymphocytes. S100A9 is also reported to induce neutrophil chemotaxis and adhesion and can increase the bactericidal activity of neutrophils by promoting phagocytosis via activation of SYK, PI3K/AKT, and ERK1/2 pathways and can induce degranulation of neutrophils by a MAPK-dependent mechanism. Thus, S100A9 is a critical protein in both normal immune function and pathophysiology, serving as a potential therapeutic target for treating inflammatory conditions. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
