General description

Monoclonal Anti-Rabbit Immunoglobulins (mouse IgG1 isotype) is derived from the RG-16 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from mice immunized with rabbit IgG. Immunoglobulins (Igs) belongs to the immunoglobulin super-family. There are five different types of Immunoglobulins. Each immunoglobulin has two heavy (H) and two light (L) chains held together by disulfide linkages. Each light comprises one variable N-terminal region and a constant C-terminal region. Heavy chain has one variable N-terminal region and three or four constant (CH1-CH4) C-terminal region. IgG is abundant immunoglobulin in human serum. The four classes of IgG can be IgG1, IgG2, IgG3, and IgG4.
Reacts with an epitope on the heavy chain of rabbit IgG, IgA and IgM, which is sensitive to reduction. No cross-reactivity with human IgG, IgA and IgM.

Specificity

Binds only rabbit Igs. Does not react with reduced rabbit Igs.

Application

Monoclonal Anti-Rabbit Immunoglobulins-Peroxidase antibody produced in mouse has been used in:
  • western blotting
  • immunoblotting
  • immunoprecipitation
  • enzyme linked immunosorbent assay (ELISA)
  • immunohistochemistry

Mouse monoclonal anti-rabbit immunoglobulins have been used for western blot assays at a dilution of 1:1000 using bovine trabecular meshwork cell lysates. The antibody can also be used for immunohistochemistry assays (1:100, using human tissue).
The mouse monoclonal anti-Rabbit IgG (heavy chain-specific) peroxidase conjugated antibody can be used as a secondary antibody for western blot analysis of protein preps from E. coli cultures at a concentration of 1:10000 diluted in PBSt for 1 hour at room temperature.

Physical form

Lyophilized from 0.01 M sodium phosphate buffer, pH 7.4, containing 1% bovine serum albumin and 0.05% MIT

Preparation Note

Prepared by the two-step glutaraldehyde method described by Avrameas, S., et al., Scand. J. Immunol., 8, Suppl. 7, 7 (1978).
