General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Specificity

Clone 28 is a mouse monoclonal antibody that detects human CD15.

Immunogen

Monocytes separated from other peripheral leukocytes on fibronectin plates.

Application

Quality Control Testing:

Evaluated by Flow Cytometry in human blood granulocytes .

Flow Cytometry Analysis: Staining of one million Whole human blood granulocytes was performed using 5 μl of a 1:1 mixture of Cat. No. CWA-1001, Anti-CD15 Antibody (28) ColorWheel® Mouse mAb and CW-PE ColorWheel® activated Phycoerythrin (PE) Dye or an equivalent amount of Mouse IgM isotype control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

Alpha-(1,3)-fucosyltransferase 4 (UniProt: P22083; also known as ELAM-1 ligand fucosyltransferase, Fucosyltransferase 4, Fucosyltransferase IV, Fuc-TIV, FucT-IV, Galactoside 3-L-fucosyltransferase, CD15) is encoded by the FUT4 (also known as ELFT, FCT3A) gene (Gene ID: 2526) in human. CD15 is a single-pass type II membrane protein of the glycosyltransferase 10 family. It contains a cytoplasmic domain (aa 1-147), a transmembrane domain (aa 148-172), and a lumenal domain (aa 173-530). It consists of a trisaccharide with the structure Gal-β(1-4)-(Fuc- (1-3))-GlcNAc and is related to another structure, sialyl-CD15 (NeuNAc- (2-3)-Gal-β(1-4)-(Fuc- (1-3))-GlcNAc; CD15s or sLex, where s refers to sialylated ), which differs only by the addition of a sialic acid linkage to the galactose in the core Lex trisaccharide. CD15 catalyzes alpha-1,3 glycosidic linkages involved in the expression of Lewis X/SSEA-1 and VIM-2 antigens. It is shown to be a distinguishing marker for human myeloid cells, and it mediates neutrophil adhesion to dendritic cells. It is also involved in neutrophil functions, such as cell-cell interactions, phagocytosis, stimulation of degranulation, and respiratory burst. It is also considered as an important marker for neoplastic Reed Sternberg cells and can be useful for the diagnosis of Hodgkin s lymphoma. (Ref.: Gadhoum, SZ., and Sackstein, R (2008). Nat. Chem. Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
