General description

The ready-to-use Alcian blue stainig kit, pH 1.0 - for the detection of strongly sulfated mucins - for microscopy, can be used for this staining in histological tissue. Alcian blue pH 1.0 is used to stain strongly sulfated mucins in human-histological specimen materials, for example histological sections of e. g. colon or bronchus. The dye Alcian blue 8 GX (Product number 1.01647) is a water-soluble phthalocyanine dye. Acidic mucins have a negative charge and bind the positively charged Alcian blue 8 GX staining dye.
A solution of hydrochloric acid and Alcian blue is used for the staining procedure. At a strongly acidic pH of 1.0, this solution suppresses the dissociation of the carboxyl groups and selectively stains only the strongly sulfated mucins and polyanions. Counterstaining takes place with a nuclear fast red/aluminium sulfate solution. The undifferentiated method (Alcian blue 8 GX solution, pH 2.5) enables only the detection of acidic glycoproteins, e. g. mucins, without distinguishing between carboxyl and sulfate groups.
