Specificity

Clone 4O5 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Bone morphogenetic protein 2 (BMP-2). It targets an epitope within the C-terminal half.

Immunogen

GST/His-tagged recombinant fragment corresponding to 101 amino acids from the C-terminal region of human BMP-2.

Application

Affinity Binding Assay: A representative lot of this antibody bound human recombinant BMP-2 with a KD of 1.8 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected BMP-2 in one million HepG2 cells.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected BMP-2 in HepG2 cells.

Anti-BMP-2, clone 4O5 ZooMAb, Cat. No. ZRB1489, is a recombinant Rabbit monoclonal antibody that specifically targets BMP-2 and is tested for use in Affinity Binding Assay, Flow Cytometry, Immunocytochemistry, and Western Blotting.

Target description

Bone morphogenetic protein 2 (UniProt: P12643; also known as BMP-2, Bone morphogenetic protein 2A, BMP-2A) is encoded by the BMP2 (also known as BMP2A) gene (Gene ID: 650) in human. BMP-2 is a member of the transforming growth factor-beta (TGF-beta) superfamily. It is synthesized as a 60-kDa precursor with a signal peptide (aa 1-23) and a propeptide (aa 24-282) which is processed in the secretory pathway to a small 18-kDa monomer. Two monomers associate to form the active homodimer that binds to its receptor. BMP-2 is shown to be abundant in lung, spleen, and colon. Lower but significant levels are also found in heart, brain, placenta, liver, skeletal muscle, kidney, pancreas, prostate, ovary, and small intestine. BMP-2 induces cartilage and bone formation and stimulates the differentiation of myoblasts into osteoblasts via the EIF2AK3-EIF2A- ATF4 pathway. BMP-2 activation of EIF2AK3 stimulates phosphorylation of EIF2A that leads to increased expression of ATF4 which plays a central role in osteoblast differentiation. It is reported to increase the colony formation of normal human bone marrow stromal preosteoblasts and induce their differentiation into cells with mature osteoblastic phenotype. BMP-2 is also known to activate osteoclast through receptor activator of nuclear kappa-B ligand (RANKL). This ZooMAb®recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

0.3 mg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
