Technically, there is no limit to the percentage of water that can be used on a bare silica column. However, exposing the column to 100% water would require several hours to re-equilibrate back to typical HILIC conditions. Nevertheless, it is advisable to use 95% water and 5% Acetonitrile to avoid dewetting the phase, unless the column is explicitly labeled as aqueous compatible. After flushing the column sufficiently, a re-equilibration of no fewer than 75 column volumes is recommended.
