Specificity

Clone 2C18 is a Rabbit recombinant monoclonal antibody that specifically detects Eukaryotic translation initiation factor 2A (eIF-2A). It targets an epitope within 23 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the C-terminal half of human Eukaryotic translation initiation factor 2A (eIF-2A).

Application

Quality Control Testing

Evaluated by Western Blotting in PC12 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected eIF2A in PC12 cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected eIF2A in mouse pancreas tissue lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected eIF2A in human pancreas tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected eIF2A in U2OS cells..

Target description

Eukaryotic translation initiation factor 2A (UniProt: Q9BY44; also known as eIF-2A, 65 kDa eukaryotic translation initiation factor 2A) is encoded by the EIF2A (also known as CDA02, MSTP004, MSTP089) gene (Gene ID: 83939) in human. eIF-2A is a widely expressed protein with higher levels observed in pancreas, heart, brain, and placenta. It functions in the early steps of protein synthesis by directing the binding of methionyl-tRNAi to 40S ribosomal subunits. It is reported to catalyze the formation of puromycin-sensitive 80 S preinitiation complexes and the poly(U)-directed synthesis of polyphenylalanine at low magnesium levels. It has seven WD repeats. Although both eIF2 and eIF-2A function in the methionyl-puromycin assays, the order of events differs between the two. In contrast to the eIF-2 complex, it binds methionyl-tRNAi to 40S subunits in a codon-dependent manner, whereas the eIF-2 complex binds methionyl-tRNAi to 40S subunits in a GTP-dependent manner. Note: The gene EIF2A should not be confused with EIF2S1 (Gene ID: 1965) that is frequently referred to as eIF2-alpha in the literature. EIF2S1 is the alpha subunit of the eIF2 translation initiator complex. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
