Description

General description

MacConkey Broth is a selective and differential liquid culture medium forcoliform bacteria. It is suitable for qualitative and quantitative detection ofcoliforms, particularly Escherichia coli, in various samples. Peptoneprovides nitrogen, carbon compounds, vitamins, and amino acids. The brothcontains lactose as a fermentable carbohydrate, bile salts to promote thegrowth of several species of intestinal bacteria and inhibit Gram‑positive organisms, and a pH indicator (Neutral red) that turns red inresponse to acid production from lactose fermentation. Gas production (whenDurham tubes are used) and/or color change are used as indicators of coliformactivity.

Application

MacConkey Broth is used as a selective enrichment and presumptive testmedium for coliforms and E. coli in water, milk, food, andpharmaceutical samples.

Preparation Note

Add 40 g to 1 litre of distilled water. Mix well and distribute into containers fitted with Durham tubes. Sterilize by autoclaving at 121°C for 15 minutes.
