General description

CDT2, also known as DTL, Denticleless protein homolog, DDB1- and CUL4-associated factor 2, Lethal(2) denticleless protein homolog, or Retinoic acid-regulated nuclear matrix-associated protein, and encoded by the gene DTL/CDT2/CDW1/DCAF2/L2DTL/RAMP, is a component of the DCX(DTL) E3 ubiquitin ligase complex and is the substrate-specific adapter for the DCX (DDB1-CUL4-X-box) E3 ubiquitin-protein ligase complex required for cell cycle control, DNA damage response and translesion DNA synthesis. The DCX(DTL) complex, also named CRL4(CDT2) complex, mediates the polyubiquitination and subsequent degradation of CDT1 and CDKN1A/p21(CIP1). CDT2 is important because degradation of CDT1 in response to DNA damage is necessary to ensure proper cell cycle regulation of DNA replication. CDT2 is localized to the nucleus primarily but it translocates to the cytoplasm during mitosis. CDT2 is expressed in multiple tissues at varying degrees with high expression in adult placenta, testis, thymus, bone marrow and fetal all fetal tissues, and low expression in adult muscle, spleen and lymph node.

Immunogen

Western Blotting Analysis: A representative lot from an independent laboratory detected CDT2 in transfected 293T cell lysates (Abbas, T., et al. (2013). Mol Cell. 49(6):1147-1158.).

Quality

Evaluated by Western Blotting in A431 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CDT2 in 10 µg of A431 cell lysate.

Target description

~85 kDa observed

Physical form

Rabbit polyclonal serum containing 0.05% sodium azide.
Unpurified

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

DDB1- and CUL4-associated factors (DCAFs) CDT2 and DCAF14 are substrate receptors for Cullin4-RING E3 ubiquitin ligase (CRL4) complexes. CDT2 is responsible for PCNA-coupled proteolysis of substrates CDT1, p21, and SET8 during S-phase of cell cycle. DCAF14 functions at stalled replication

The timely and precise duplication of cellular DNA is essential for maintaining genome integrity and is thus tightly-regulated. During mitosis and G1, the Origin Recognition Complex (ORC) binds to future replication origins, coordinating with multiple factors to load the minichromosome
