General description

Anti-Doublecortin, Cat. No. AB2253, is a guinea pig polyclonal antibody that detects Doublecortin and is tested for use in Immunohistochemistry (Paraffin) and Immunofluorescence, and Western Blotting.
Target molecular weight ~42 kDa observed;40.61 kDa calculated. Uncharacterized bands may be observed in some lysate(s).
~45 kDa observed; 40.57 kDa calculated. Uncharacterized bands may be observed in some lysate(s).

Immunogen

Epitope: C-Terminus
Recombinant Peptide.

Application

Anti-Doublecortin Antibody has been used in immunohistochemistry and immunofluorescence staining.

Biochem/physiol Actions

Doublecortin (DCX) is identified as an important marker for neurogenesis. It serves as a valuable tool for studying and comparing the process of neurogenesis across different species.[1]
Reactivity with other species has not been determined.
This Guinea pig polyclonal antibody detects doublecortin in murine and rat species. It targets an epitope with in 17 amino acids from the C-terminal region.

Physical form

GST Depletion
GST-Depleted Guinea Pig Sera with 0.05% Sodium Azide

Preparation Note

Stable for 1 year at -20°C from date of receipt. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Analysis Note

Control
Rat Brain lysate.
Evaluated by Western Blotting in rat brain tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Doublecortin in rat brain tissue lysate.

Questions

  1. Is it possible to obtain a small aliquot of specific antibodies for testing purposes to determine their suitability for staining Doublecortin (DCX) in immunohistochemistry on free-floating mouse brain tissue fixed with PFA? The antibodies of interest are the guinea pig polyclonal and the goat polyclonal.

    1 answer
    1. After verification, an anti-doublecortin antibody recommended for free-floating application on mouse brain tissue is not currently available. However, a citation on AB2253 for free-floating application on mouse brain tissue was found and may be of interest for reference.

    No longer works

Our lab used this product because it previously stained neuroblasts in the mouse brain (1:500 on 4% PFA fixed, frozen tissue sections) very reliably. Unfortunately, the recent batches of this product no longer stain any neuroblasts in the mouse brain.

    1. Response from MilliporeSigma:

We are sorry to hear your recent lot of AB2253 has not been staining consistent to past lots. This product is suitable for IHC with mouse primary neurons. Optimal dilutions need to be determined by the end user for each lot.

We would like to discuss this further with you so we can investigate the lot. To contact us for additional technical support, please visit https://www.sigmaaldrich.com/US/en/support/customer-support to submit a Product Technical Inquiry.
