Poly(deoxyinosinic-deoxycytidylic) acid (Poly(dI-dC) • Poly(dI-dC)) is an alternating copolymer used as a DNA substrate for evaluation of DNA methytransfeases, such as DNA-methyltransferase 1 and as double-stranded DNA model for conformational studies of DNA structure dynamics and drug, small molecule, interactions.
Poly(deoxyinosinic-deoxycytidylic) acid sodium salt has been used in a study that synthesized and characterized bis(2-(pyrimidin-2-γl)ethoxy)alkanes and their pharmacological activity. Poly(deoxyinosinic-deoxycytidylic) acid sodium salt has also been used in a study that investigated the growth of calcium carbonate films on LB/LbL matrices.
Poly(deoxyinosinic-deoxycytidylic) acid sodium salt has been used in the electrophoretic mobility shift assay in p65 protein[1] and neuro-2A protein lysate.[2] It has also been used as a substrate in the protein arginine methyltransferase 1 (PRMT1) and DNA methyltransferase 1 (DNMT1) selectivity assay.[3]

Poly(2′-deoxyinosinic-2′-deoxycytidylic acid), poly(dI-dC) is a synthetic DNA substrate.[4] At high salt, Poly(dI-dC) exists in left handed helical conformation and reverts to right-handed form upon decreasing salt.[5] It is used in electrophoretic mobility shift assay (EMSA).[6]

    1. The polymer size of this material varies from lot to lot. The typical low range is 150 to 550 base pairs and the typical high range is 3000 to 5000 base pairs. The bulk of the product ranges from 1200 to 3000 base pairs. The molecular weight of a single Poly(dI-dC) base pair is 639.41 g/mol, resulting in an approximate molecular weight range of 767.2 kDa – 1,918.2 kDa for most of the compound. The unit per milligram value may range from 8 – 14 units per milligram and the vial content may range from 5.0 – 7.5 units.     1. This product is soluble in water or neutral buffer. Solubility is no longer considered part of the specifications, but it was previously reported to be soluble at 20 mg/mL in 0.1 M Tris-HCl, pH 8.8. Heat is not necessary to solubilize the product; dissolving in an appropriate solvent is the recommended method for resuspension.
